An Experimental Study of the Effect of Halothane Anesthesia on the Liver Function in



Abstract
The present study were performed to determine the effects of the halothane anesthesia on the liver function in Sprague-Dawley rats pretreated with isoniazid. Ninety six male Sprague-Dawley rats were divided into four groups. Saline(2ml/kg, in
group
I)
or isoniazid(50mg/kg, in group II) was injected intraperitoneally for 7 days, and then exposed to sham anesthesia 24 hours after the last treatment. Saline(2ml/kg, in group III) or isoniazid (50mg/kg, in group IV) was injected intraperitoneally
for
7
days, and then exposed to 1% halothane(N2O-O2 : 3L/min-3L/min) 24 hours after the last treatment. Blood sampling was done before any treatment through tail vein. Intracardiac puncture for blood sampling was done after intraperitoneal injection of
pentobarbital(50mg/kg) and hepatectomy was done to get the tissue sample for light microscopic examination 24 hour after exposure to air or halothane in one-half of the rats from each group; the remaining half of the rats from each group were
examined
96 hours after exposure to air or halothane. Biochemical studies included liver function variable (protein, albumin, total bilirubin, direct bilirubin, alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase).

@EN The histologic evidence of hepatic damage was more severe in group II and group IV than I and Group III respectively 24 hours after exposure to air or halothane. In group IV, the histologic score revealed more severe hepatic damage than group
II.
2. The histologic evidence of the hepatic damage was more severe in group IV 96 hours after exposure halothane.
3. The biochemical study showed no significant differences among the groups.
The above results suggest that isoniazind itself can produce liver injury and exposure to halothane may aggravate the isoniazid induced hepatic injury.
